    Abstract:

    To study mouse oocytes in vitro maturation and fertilization in different culture. Methods Compare the maturation and fertilization rate in vitro of mouse oocytes, which were culture in FSH, Insulin or BSA, and were fertilized in Whitter's medium. Results 1.IVM/IVF rate of Do (81.4%, 31.0%) were higher than those of COC (48.6%, 27.1%). 2.Mouse oocytes cultured with FSH, insulin and BSA, the IVM rate were 77.9%, 82.3% and 60.7%, the IVF rate were 77.2%, 72.6% and 26.7%, the 2-cell rate were 49.2%, 34.2% and 10.0% respectly. The group cultured with insulin (I-Group) have the highest rate of IVM, but after fertilization, fertilized oocytes and 2-cell embryos of I-Group are fewer than those cultured with FSH. 3.two groups cultured with BSA have lower rate in vitro fertilization rate (26.7%, 25.8%). 4.Fertilization and 2-cell rate in those oocytes with polar body 1 (85.9%, 22.4%) were higher than those of GV-stage oocytes (71.1%, 12.9%). Conclusions 1.In vitro maturation and fertilization rate of Do were significantly higher than those of COC (P m<0.01; P f<0.05). 2.The maturation and fertilization rate were improved when the mouse oocytes were cultured in FSH and Insulin mediums. 3.BSA affected the fertilization rate of mouse oocytes significantly. 4.The IVF rate and 2-cell rate of oocytes with PbI are higher than those of GV-stage oocytes (P f<0.05; P 2-cell <0.05).
