    目的 构建大鼠胃炎癌转化模型,探索新的胃癌早期血清学诊断指标。方法 采用N-甲基-N'-硝基-N'-亚硝基胍饮水(MNNG)协同高盐饲料喂养的方法诱导大鼠胃炎癌转化模型。核磁共振成像及组织病理学观察大鼠胃部恶化过程。qRT-PCR检测不同时间点大鼠血清miR-17-5p和miR-374-5p表达水平。结果 MNNG饮水协同高盐饲料喂养成功诱导大鼠胃炎癌转化,经历萎缩性胃炎和不典型增生的过程。血清中miR-17-5p和miR-374-5p的表达水平与大鼠胃部恶化程度正相关。结论MNNG饮水协同高盐饲料喂养构建的大鼠胃炎癌转化模型为研究胃癌发病机理提供了良好的动物模型,血清miR-17-5p和miR-374-5p是潜在的胃癌早期无创诊断的指标。

    Abstract:

    Objective The transformation from gastritis to cancer model of rat was constructed, and the new serological diagnosis index of the gastric cancer was explored. Methods N-methyl-N'-nitro-N'-nitrosoguanidine (MNNG) drinking water and high salt feed were used to induce gastric cancer in rats. Dynamic observation of gastric deterioration in rats was operated by nuclear magnetic resonance imaging and histopathology. The expression of miR-17-5p and miR-374-5p in the serum of rats was detected by qRT-PCR in different time points. Results MNNG cooperated with high salt diet could induce gastric degeneration, through the atrophic gastritis and atypical hyperplasia process. The expression level of miR-17-5p and miR-374-5p in the serum was positively correlated with the degree of gastric degeneration in rats. Conclusion the rat gastritis cancer transformation model constructed by MNNG drinking water combined with high salt feed provides a good animal model for studying the pathogenesis of gastric cancer. Serum miR-17-5p and miR-374-5p are potential markers for non-invasive diagnosis of gastric cancer at the early stage.
