The Gradual Changes of Myocardial Fibrosis and AGEs/RAGE Levels of Diabetic Rats

Abstract:
      【ABSTRACT】 Objective In order to investigate the gradual changes of myocardial fibrosis and myocardial AGEs/RAGE levels of type Ⅱ diabetes, the authors established the rats model for type Ⅱ diabetes. Methods Male SD rats werefed high-fat fodder for 6 weeks, and then they were injected a small dose of streptozotocin (STZ, 30mg/kg) after overnight fasting 12h. The random blood GLU was detected in the 3th and 7th day respectively after STZ injection. When the rats of which the random blood GLU concentration was greater than or equal to 16.7 mmol/L in two times were successful rats’ model of type Ⅱ diabetes. Six rats of group C and six rats of group D with fasting 12h were killed after intraperitoneal anesthesia at the 4th, 8th and 12th week after the type Ⅱ diabetic rats successfully established. The blood was used for FBG, insulin detection. A portion of the left ventricle was used for Masson staining and immunohistochemical detection of RAGE, and the other portion of the left ventricle was used for the content detection of AGEs and Hyp. Results The FBG level of group D were significantly higher than them of group C at the same week. The body weight of group D were significantly lower than them of group C at the same week. Blood insulin of group D at the 12th week were significantly higher than those of group C at the 12th week and group D at the 4th week. Myocardial Hyp, CVF, AGEs and RAGE of group D were significantly higher than those of group C at the 8th, 12th week. The AGEs/RAGE level of myocardium in type Ⅱ diabetic rats was positively correlated with CVF. Conclusion The model of type Ⅱ diabetes were successfully established through intraperitoneal injecting a low-dose streptozotocin to SD rats that had been fed high-fat fodder. The rats of type Ⅱ diabetes demonstrated myocardial fibrosis at the 12th week after the diabetic rats’ model successfully established. With the further development of type Ⅱ diabetes, myocardial fibrosis, the content of myocardial AGEs, RAGE protein expression of the diabetic rats had an upward trend. The level of AGEs/RAGE was positively correlated with the degree of myocardial fibrosis of type Ⅱ diabetic rats.
