The Establishment of the Immuno-Reconstituted hu-PBL-SCID Mice and the Effects of Their Human Lymphocytes on Human Lung Carcinoma Metastasis

    Abstract:

    Human peripheral blood lymphocytes (hu-PBL), were transferred to SCID(severe combined immunodeficient) mice by intraperitoneal injection, thus hu-PBL-SCID mice model was successfully established. Significant levels of human immunoglobulin were detected in sera of hu-PBL-SCID mice 4 weeks after engraftment. 8 kinds of immunonological phenotype of human lymphocyte were detected in the spleens of hu-PBL-SCID mice by flow cytometry 4 weeks after engraftment. These data demonstrated that proliferation or reconstitution of human lymphocytes appeared in the SCID mice. It suggested that and there was no difference of metastases between hu-PBL-SCID mice and unreconstituted SCID mice receiving transplanted human lung carcinoma PLA-801DL. The human lymphocytes in the hu-PBL-SCID mice have no significant antitumor effect.
